Synthetic biology – fungus

fungus

The word fungus does not conjure up pleasant memories to most. This has negative implications though mushrooms are a group of mycelial aggregates too. They form an important degradation pathway in nature and are important for recycling much of the heavy plant material. Fungus has not been a major part of producing anything relevant but a company called Bolt Threads is aiming to use it to manufacture leather equivalent. The market for leather is huge and is derived as a by-product of the animal industry. In some cases, it is not necessarily true – snake skin is harvested from the snake by killing the snake for the skin and not for the meat as in other industries.
Bolt Threads aims to grow mycelium and then cure it into leather. This is something unique and promises to be sustainable way to produce leather, rather than animals or synthetically from oil based economy.
The images on their website are amazing. It shows a very leather like material that is made from fungi in large containers!
Though they do not disclose much on their website, it will be interesting to see what kind of synthetic biology process went into creating genes/cytoplasmic/enzymatic processes that produces proteins or other substances which behave and feel like leather.

    TDP43

TDP43 (a nuclear DNA-RNA binding protein) is involved in splicing, RNA transport and stability in neurons. It was first identified as a pyrimidine rich TAR DNA sequence in HIV type 1 gene to modulate HIV gene expression. In 95% of the ALS patients, TDP43 is abnormal located within the cell and may also be aggregated,…

  • Spider robot molded cheaply

Cheap robots by the millions Typically it costs many hundreds of dollars to make a robot that can do something substantial. However, researchers at the Fraunhofer institute have created molded or stamped robots that can be created very cheaply – they imagine them to be like disposable gloves. Make them, use them and then throw…

Small Biotechs: Diagonal Tx: Clustering antibodies that mimic the action of the ligand and bypass the need for the ligand and receptor. This mutation that is created makes standard AI models not useful and so need a new method. This restores new ALK1 signaling in Hereditary Hemorrhagic Telangiectasia. It also treates LoF mutations in ALK1…

  • Vitamin K2

It has been traditionally thought that for bone health, sufficient calcium is required. However, it is just not calcium. A vitamin called Vitamin K1 (phylloquinone) and K2 (menaquinone) in combination with Vitamin D2 and calcium is important for activating a protein called osteocalcin which binds to calcium to build bones. Osteocalcin is also involved in…

    Fitness landscape for antibodies (Flab2)

There have been many AI models to predict the structure of proteins, especially antibodies. There are several key aspects to developing antibodies as drugs beyond binding to the target of interest. These are : thermostability, expression, aggregation, binding affinity, pharmacokinetics, polyreactivity and immunogenicity. There have been many models available such as IgLM, ProGen2, Chai-1, ESM2,…
